How to use this calculator
Enter the total cells counted, the number of large squares, the dilution factor and your sample volume. Cells per mL and total cells are calculated.
Formula
cells/mL = (cells counted ÷ squares) × dilution × 10⁴
Example
180 cells in 4 squares, 1:1 trypan blue, 10 mL sample:
- Cells/mL = 45 × 2 × 10⁴ = 9.0 × 10⁵
- Total = 9.0 × 10⁶ cells
Frequently asked questions
How do I calculate cells per mL with a hemocytometer?
Cells/mL = average count per large square × dilution factor × 10⁴. Each large square holds 0.1 µL (10⁻⁴ mL).
How many squares should I count?
Usually the four corner squares (and sometimes the centre), aiming for 100–300 cells in total for accuracy.
What dilution does trypan blue add?
Mixing equal volumes of cells and trypan blue gives a dilution factor of 2.
